3种方法测定血液中微量白细胞计数效果的比较研究

 提要 : 目的 比较牛鲍式计数板、血球计数分析仪和Nageotte 血 细胞计数 板3 种白 细胞计数 法测定血液中微量白细胞的效果。方法 将1 份血液标本重复用3 种方法计数,得出标准计数值,并将此标本稀释至不同浓度,以3 种计数法分别测定这些样品中白细胞的含量,比较测试值与期望值。同时,用这3 种方法检测了30 份过滤白细胞的血样中的微量白细胞。结果 牛鲍式计数板、血球计数分析仪在白细胞< 200 ×106/ L 时,牛鲍式计数板测试值为“0”, 血球计数分析仪测试值为200 ×106/ L ,无法准确反映低 含量样品中白细胞的真实结果,Nageotte 血细胞计数法则可准确测定血液中> 112 ×106/ L 的白细胞。结论 测定血液中微量白细胞宜采用Nageotte 血细胞计数板方法。    关键词 : 白细胞计数;血液制剂;Nageotte 血细胞计数法   中图法分类号: R457    文献标识码: A Comparison of 3 methods for counting white blood cells in diluted blood samples ZENGJie , CHENGXiao2ling , Wang Ze2rong , REN De2hui , LINGWu2cun , ZHAO Shu2ming(Department of Transfusion , Southwest Hospital , Third Mili2 tary Medical University , Chongqing 400038 , China)   Abstract : Objective  To compare the results of WBC counting in blood with small amount of WBC by Nageotte hemocytometry , Neubauer chamber method and automatic particulate analysis. Methods  A blood sample of 5 ml was counted with routine method for 3 times to obtain its WBC count as a standard value , and then diluted with saline to different levels. All of those blood samples were counted for WBC concentrations by microscopic counting with a Nageotte hemocytometer (Haucser , USA) , a Neubauer chamber , and an automatic particulate analyzer (F - 820 Symex , Japan) . The accuracy of each method was compared between the counting results and the expected values. The apparatuses were also used to deter2mine WBC counts of 30 blood samples after leukocyte filtration. Re sults  When the blood samples with WBC < 200 ×106/ L , Neubauer chamber got 0/ L , and the automatic particulate analyzer showed 200×106/ L , which were far away from the theoretical values. A good accuracy was obtained by Nageotte hemocytometer in the samples with WBC counts between 1. 2×106 to 200 ×106/ L. Compared with the other 2 methods , Nageotte hemo2 cytometry showed more accurate results in the WBC counting of red blood cell concentrations. Conclusion  These results suggest that Nageotte hemocytometry is a good method for counting micro2WBC in blood components , better than the other 2 methods.
